Abstract

Medicinal plants remain important sources of structurally diverse secondary metabolites for pharmaceutical discovery, but the quality of an extract depends strongly on sample preparation, solvent polarity, extraction conditions, fractionation and analytical screening. This methodology paper presents a reproducible workflow for extraction, isolation and phytochemical screening of medicinal plant material, using Moringa oleifera leaves as a representative matrix. Shade-dried powder is extracted with 70% ethanol by controlled maceration with ultrasonic assistance, concentrated under reduced pressure and sequentially partitioned into n-hexane, ethyl acetate, n-butanol and aqueous fractions. Thin-layer chromatography (TLC) is used to select chemically distinct fractions, followed by silica-gel column chromatography for isolation of enriched subfractions. Qualitative tests are performed for alkaloids, flavonoids, phenolics/tannins, saponins, terpenoids, cardiac glycosides and reducing sugars, while total phenolic and total flavonoid contents provide quantitative indices. The proposed design includes extraction yield, chromatographic profile and phytochemical class distribution as primary responses. An illustrative validation dataset demonstrates how the method can identify a hydroethanolic extract rich in polar and semipolar constituents and how ethyl-acetate fractions can concentrate phenolic/flavonoid components. The procedure integrates conventional pharmacognostic methods with solvent-efficient extraction and chromatographic isolation, producing a practical platform for subsequent bioactivity-guided pharmaceutical studies.
